The characterization of RNA–protein complexes (RNPs) is a difficult but increasingly important problem in modern biology. Mango-tagged RNP complexes can easily be purified from cell extracts. Using streptavidin solid support and TO1-3PEG-Desthiobiotin (TO1-DTB) allows the recovery of RNP complexes in their native state by the addition of free biotin. Thus a Mango tagging strategy enables not only RNA imaging of live or fixed cells, but also the rapid and scalable purification of complex RNP complexes
